Hello,
Many including me have been asking for such a list, and I recently made all my heroes 70 so I'll try to make a list.
Please inform me if you think anything is wrong or missing.
Edit: Got Neil & Nixaes
Legend
DPS - Damage dealer
Tank - A hero that can take a lot of damage
AoE - Area effect
Score meanings:
5/5: You must use him if you have him.
4/5: There's often a place for him.
3/5: You can use him ocassionally.
2/5: He can be used as a replacement.
1/5: Don't use him.
Tier 1: Legendary heroes
Zorak: Zorak is arguably the best hero since he is useful in all game modes. He is an ordinary 3 mobility melee physical DPS, but his ability to lock down the skills of a hero (often Helena or enemy's Zorak), inhibit healing on that hero, and kill him/her in round 2, combined with his stealth and the ability to block paths of enemy non-fliers is crucial. Everybody uses him on defense and if you don't have him, you may lose your Helena or another hero, so you must have him, both in PVP and Sky Arena. He is a little squishy but he knows how to get out of harms way thanks to his teleport ult. Also he can't be counter attacked and has a lot of dodge so he doesn't fall that easily. Also he is stun immune which means you can use him against the guild boss, especially when he has 0 or 1 tentacles left. He is also useful in a few heroic levels.

Nixaes: Our new hero is amazing at several stuff. First off even his nerfed AoE damage is on par with Elemix, and he is much more reliable, as he is immune to everything. Secondly with 6 mobility he dives into the enemy in PVP, which normally is a bad thing but along with his egg he occupies the enemy longer than any unit before dying. Third, while he can't be healed, he is somewhat reliable in the Sky arena because you can just let him go egg and protect him and he comes back at full health, even if the match ends before he breaks the egg. Third, his 1st skill rips apart when he is on low health making him a surprise assassin. You don't necessarily need him but for me he is a better version of Elemix. Finally he can do some odd stuff and help you pass some heroic levels.

Helena: The only main healer in the game, Helena is the most crucial hero to have in your team in the Sky Arena. Don't let the healing abilities of Aqua or Trinity fool you. Helena's 3 of 4 skills is on healing. She heals single target, her ult heals AoE, and she can self heal. She's very squishy at the beginning but when fully equipped, she can take more punches than most ranged heroes. Her damage is crap and if you're relying on her damage, you're in trouble.

Centaur: Centaur is the king of crowd control and the best tank in the game. He can take a lot of punches before he falls, and his turn 1, 4, 7 stun + his ult's mass stun is what saves your neck in the sky arena. His turn 1 mobility gets him into the heat of the battle before any other tank, which is a great advantage. His CC also makes him a must have for heroics. His only downsides are, 1- he can't deal damage. 2- He is not a flyer so he can sometimes struggle with obstacles. Still, he is the most mobile of all tanks. Also there are only 2 stun immune heroes (Zorak & Igor) so his CC actually works. One more thing; he's low counterattack damage renders him a viable target for enemy AI which helps him protect your weak units.

Pixie: If Centaur is the king, Pixie is the queen of crowd control. She sleeps a random hero (lasts 1 turn) before turn 1, 4, 7 and her ult randomly sleeps, petrifies, or lowers the attack of a hero (this petrify and sleep can last 2 to 3 turns). Of course this is not as reliable as Centaur's stun but she can use her ult once every 2 turns. Also the only sleep immune hero is Farand so her CC also works. Her damage is quite good and she is one of the heroes that can't be counter attacked. The downside is she is too squishy and you can't use her for long without healing.

Dracula: Dracula is the only Tier 1 non-living. (Note: undeads, elementals and demons are often unusable because they can't be healed). He is tougher than most flyers, he hits as hard as others, can't be counter attacked and he can heal himself very well. Just try not to get him focused on, but his mobility helps him avoid focus. His big downside is he has no CC, but his ult can kill, so you can use him safely. I also use him in guild boss when he has no tentacles left (because you need a 4 mobility hero with high damage).

Gladius: Gladius is a must have only because of the guild boss. He seems to be spesificly designed for the encounter. Deals constant damage with wingflap, strikes 2 targets back to back with normal attack & counter, ult deals massive AoE damage. None of these are very useful in PVP or sky arena, but he is the single most top damage dealer in guild boss and he is reliable doing that (unlike Elemix). His only CC is wing flap's pushback which is crap.

Kamus: Kamus is in this tier for the same reason as Gladius. This tanky slow guy can deal a lot of damage (especially with his ult), but he shines because of his permanent defense reduction. This helps to nullify the high defense of the guild boss (1 tentacle) which you focus your ranged and gain damage with everyone. He is tough so you can also use him in PVP. Has no crowd control.

Elemix: Elemix is an odd hero. His normal & counter attack deals all around damage, which is awesome for guild boss but he can be stunned which renders him unreliable. If he doesn't get stunned the whole fight, he will help you break the server record (he and Nixaes share the top damage dealers at this point, but Nixaes doesn't get stunned while Elemix does). His ult is the single best ult in the game, stops the whole enemy team and no hero is immune. But this has nothing to do with the guild boss, and he is way too squishy for a melee for sky arena (he is elemental so can't be healed). He also sends back the melee with his normal attack which can render Zorak useless. If you have him, use him in Sky Arena for a few rounds but he will eventually die.
However in Arena PVP he can sometimes do amazing stuff. Still not very reliable though.

Pei: He is tough, has regen, ult deals a lot of damage, but the thing that makes him shine is his battle roar which amplifies all the team's damage. This means 20k+ damage for the team. He has to be fully geared and starred to be able to kill the fishes with counter attack (which helps him use his ult twice). Has no CC.

Hyperia: She's not very reliable but in PVP she can be the deciding factor with her amazing dodge, not letting anyone use their 1st skill, and good damage. But she's so squishy so she may not do any of these. She regens, so she can be used in the sky arena too.

Tier 2: Very Good heroes
Cyclops: Toughest ranged. The enemy will not kill him without focusing for 2 rounds and his ult doesn't let them do that. His passive bomb also helps damage the guild boss. Interestingly his damage is pretty awesome too, even better than Pei.

Isha: Her ult is the biggest single target damage in the game. Can replace Gladius in guild boss fight if you or a guildie doesn't have him. She is so slow and squishy, and has no CC.

Grump: The toughest of tanks in the game, Grump's auto taunt is one of the biggest Zorak pacifiers. Use him in a low mobility team and he'll prevent the enemy from winning under 10 turns in PVP. His low damage is somewhat remedied with cooperation if used together with Alex.

Aqua: The second best healer in the game, Aqua can't heal herself, but has an amazing round 1 crowd control. Elemental means not very viable in Sky Arena where healers actually shine. Still in Live Arena she rocks major.

Igor: Poor man's Nixaes, Igor is stun immune so we can say he is designed for guild boss. He deals a little less damage but he is alright. He's not very viable for anywhere else but although he can't be healed, he can eat corpses (ewww) to regen is HP.

Farand: A good tank, just not as tough as Centaur & Grump. He lowers the defense of other heroes and can deal some damage later on. Being a flyer helps, being undead doesn't. He can be used to lower defense of the guild boss instead of Kamus, because he can lower the defenses of multiple tentacles at once.

Tier 3: Good heroes
Merlin: A mediocre damage dealer, is extremely squishy but his round 1 protection for the whole team is very valuable. Can help with guild boss if 1 or more tentacles are down.

Isaac: He is a fine single target damage dealing ranged. His normal attack and ult is effective on killing even the toughest tanks, not to mention he can take out ranged on turn 1 in the Sky Arena if he starts with full energy. Only CC is his melee pushback. He is demon but can self heal much like Dracula. Also he has no range, which means he deals full damage from melee to 10 range. Using him with her sister doesn't often help him survive.

Alexander: He is a little to squishy for a tank but his damage is higher than most either. Has a huge self heal on turns 3,6,9 but if focused he dies unlike Grump & Centaur. His best side is coop where he shines with Grump.

Trinity: Third best healer, Trinity can group heal and has crowd control. Very squishy so being melee doesn't really help. I find him especially usable in the Live arena.

Pegasus: Her damage ROCKS, her 1st skill can kill, but she's sooo squishy that she can't do any of these. The worst VIP hero. Some say she's good in heroics but I don't know why one need a hero without cc in heroics. Also in guild boss, her 1st skill doesn't do extra damage.
